The diagnosis of potato virus Yn, directly from tubers of secondarity infected potato plants, was carried out by the following method:
1.- Cutting off a 4 mm piece from each end of the tubers.
2.- Storage of the cut tubers from 10 to 14 days at 20°C.
3.- Inoculation on Solanum demissum A6 leaves, with the exudate of the new cut at the sprout end. The inoculated leaves were stored in a wet chamber at 24°C. and a ligth intensity of 1.500 lux
The infection of potato virus Yn was observed in all the tested tubers. The comparative couming of the local lesion produced on the inoculated leaves, proved that the concentration of the virus at the sprout end of the tubers was similar to that of the leaves of the secondarily infected potato plants.
